We set out to generate a test a novel method of delivering our new generation of genetically encoded calcium indicators systemically to transgenic mice in vivo. We have so far cloned generated and amplified two recombinant adeno-associated viruses (serotype 9) that will express cytoplasmic and myofilament localized RGECO indicator in the heart of mice once administered via tail vein injection. We have begun in vivo testing to determine titre and dosing time course requirements. Once established this will allow a number of inter and intra CRE collaborations to be fostered to assess calcium flux in a wide number of cardiac disease pathologies.    

Thin pump priming application has enabled the development of a novel tool kit of in vivo-deliverable calcium indicators that will be important for detailed mechanistic studies by Oxford CRE researches on a large variety of disease models and pathologies such as heart failure, hypertrophy, or arrhythmia.
