Probing the stability of dynamic RNA multi protein complexes

The fundamental biological process of translation of m-RNA on the ribosome was examined. Samples were introduced using nanoflow electrospray methods from borosilicate glass needles, and all spectra were recorded on a modified Q-ToF mass spectrometer adapted for the transmission and analysis of higher molecular weight complexes. Indroducing E. coli ribosomes from different solution conditions revealed the propensity for certain regions of the ribosome to become unstable. The results show that the extent of protein RNA interactions are the major effect in governing the dissociation of proteins from the ribosomes.
