Oroboros O2k FluoRespirometer
- The Oroboros oxygraphy is a high resolution respirometer for oxygen consumption measurements in isolated mitochondria, permeabilized or intact cells, permeabilized muscle fibres or tissue homogenates (fresh and frozen).
- Individual electron transport chain complexes, max respiration rates, leak respiration and mitochondrial coupling can be assessed with a range of substrates and inhibitors (including cell permeable versions).
- The Oroboros also has an integrated Fluorimeter, which can be used to detect hydrogen peroxide production via Amplex red, calcium concentration via calcium green, ATP production via magnesium green or mitochondrial membrane potential via safranin, TMRM or TPP+
- The amount of biological material needed is roughly >50 ug mitochondrial protein equivalent, 10^6 cells or 5 mg tissue. Two samples can be run in parallel, and a typica comprehensive run with a range of substrates and inhibitors (SUIT protocol) lasts 60-90 minutes.
